Or perhaps your refering to off screen references, in which case Captain Pike was the first to use the term. Captain Pike served as Captain of the Enterprise before Kirk, as I'm sure you know, and his second in command was Number One. The female character was not given a proper name, however Roddenberry had intented her to be a regular character on the show.
I'm gonna wait for a confirmation on that one before I ask my questions, just because I'm not usre...also, I've got some pretty intense questions...

Capt. christopher pike was the answer I was looking for.
Officially "Number One" was coined in the OS episode "Menagerie" though it was really the first pilot, "the Cage".
Go ahead and try and challenge fellow trekkies (or Trekkers)

Time to bring this back.
There were four ships not just one, Federation starships Lexington, Potemkin, Excalibur, and Hood.
